Prospective Subunit Nanovaccine against < em > Mycobacterium tuberculosis < /em > Infection ─Cubosome Lipid Nanocarriers of Cord Factor, Trehalose 6,6' Dimycolate
ACS Appl Mater Interfaces. 2023 Jun 1. doi: 10.1021/acsami.3c04063. Online ahead of print.ABSTRACTAn improved vaccine is urgently needed to replace the now more than 100-year-old Bacillus Calmette-Guérin (BCG) vaccine against tuberculosis (TB) disease, which represents a significant burden on global public health. Mycolic acid, or cord factor trehalose 6,6' dimycolate (TDM), a lipid component abundant in the cell wall of the pathogen Mycobacterium tuberculosis (MTB), has been shown to have strong immunostimulatory activity but remains underexplored due to its high toxicity and poor solubility. Herein, we employed a novel ...

The Application of New Approach Methodologies in Respiratory Disease Research: Their Role in Improving Translational Medicine from Bench to Bedside
We describe their potential uses for efficacy and toxicity assessments, to optimise the drug development process and reduce the high failure rates in clinical trials.PMID:37067473 | DOI:10.1177/02611929231170392

Highlights into historical and current immune interventions for cancer
Int Immunopharmacol. 2023 Feb 27;117:109882. doi: 10.1016/j.intimp.2023.109882. Online ahead of print.ABSTRACTImmunotherapy is an additional pillar when combined with traditional standards of care such as chemotherapy, radiotherapy, and surgery for cancer patients. It has revolutionized cancer treatment and rejuvenated the field of tumor immunology. Several types of immunotherapies, including adoptive cellular therapy (ACT) and checkpoint inhibitors (CPIs), can induce durable clinical responses. However, their efficacies vary, and only subsets of cancer patients benefit from their use. In this review, we address three goal...

Indirect effects of cytomegalovirus infection: Implications for vaccine development
Rev Med Virol. 2022 Nov 15:e2405. doi: 10.1002/rmv.2405. Online ahead of print.ABSTRACTDevelopment of a cytomegalovirus (CMV) vaccine is a high priority due to its significant global impact-contributing to mortality in immunosuppressed individuals, neurodevelopmental delay in infected neonates and non-genetic sensorineural hearing loss. The impact of CMV on the general population has been less well studied; however, a wide range of evidence indicates that CMV may increase the risk of atherosclerosis, cancer, immunosenescence, and progression of tuberculosis (TB) and human immunodeficiency virus. Due to the high seroprevale...

Implications of the non-specific effect induced by Bacillus Calmette-Guerin (BCG) vaccine on vaccine recommendations
CONCLUSIONS: Despite evidence obtained from in vitro studies, and some epidemiological and clinical evidence, more robust evidence of in vivo efficacy is still needed to justify the use of BCG in clinical practice, in addition to what is recommended by the National Immunization Program for TB prevention and bladder cancer treatment.PMID:36309066 | DOI:10.1016/j.jped.2022.09.002
